Osmogen mediated hollow microparticles fabrication for peptide encapsulation
and delivery
abstract
Bioactive molecules such as peptides and proteins have
significant therapeutic potential. However, their therapeutic ability is
limited by their low systemic stability, short half-life, and subsequent loss
of bioactivity. In order to increase their stability, drug efficacy, and to
achieve a sustained release profile, encapsulation with
poly(lactic-co-glycolic acid) (PLGA), is carried out.
PLGA hollow microparticles were fabricated by a novel osmogen mediated
solvent emulsion evaporation technique with the aim of minimizing the
creation of a localized acidic microenvironment, which causes peptide
denaturation. The effects of various process parameters on the morphology of
the hollow microparticles were studied and characterized by scanning electron
microscope (SEM). Thereafter, these hollow microparticles were loaded with
the peptide, Glucagon-like peptide-1 (GLP-1), and its subsequent
encapsulation efficiency and in vitro release behaviour were analyzed.
